TITLE: "EM to X-ray: Case Studies"
ABSTRACT: Multi-component molecular complexes are now being tackled by structural biology, bringing X-ray crystallography into the realm of electron microscopic (EM) studies. X-ray structure determination can utilize a low-resolution EM image for molecular replacement (MR) followed by phase extension to higher resolution by density modification. Case studies will be presented that use EM images as models for crystal structure solution by molecular replacement with AMoRe, MolRep and Phaser. Considerations for the EM data, such as manipulating the EM image and the effects of EM magnification error, the dependence of the MR solution on X-ray data completeness and intensity distribution, and the effects of MR positional/rotational errors on phase extension will be discussed.
