TITLE: "Molecular Replacement and structure completion in PHENIX"

ABSTRACT: Approximately two-thirds of the new structures deposited in the Protein Data Bank are phased using molecular replacement or difference Fourier methods. Although often easier to achieve than phasing using isomorphous replacement or anomalous diffraction, the molecular replacement method suffers from the problem of model bias. Robust methods for rebuilding models, especially at medium to low resolution, are required to minimize the chances of errors in the final model. PHENIX achieves this using a combination of maximum likelihood molecular replacement methods (PHASER), statistical density modification and automated model (re)building (RESOLVE), and automated structure refinement (phenix.refine). Results using this approach will be presented.
